Separation & Purification
The TFF system provides a rapid and efficient method for separating and purifying biomolecules. It utilizes fluid filtration, which can be either microfiltration or ultrafiltration, depending on the type of membranes used.
Parallel Filtration
The filtration process occurs in parallel to the membrane and involves the application of a pressure difference. This setup allows smaller volume components to pass through the filter (permeate), while larger volume components continue along the membrane (retentate).
Permeate and Retentate
The product that successfully passes through the filter is referred to as permeate. It is the purified fraction that has undergone separation from the biomolecules.
Up to 0.7 m²
The TFF equipment comprises two primary modules. The first module is the filtration assembly, which includes a single-use eBAG® plastic tank with a volume ranging from 2 to 5 liters. This tank holds the product that requires filtration. The assembly also contains a filtration membrane with a surface area of up to 0.7 m².

The TFF system enables a fast and efficient process for separation and purification of biomolecules. Fluid filtration can be either microfiltration or ultrafiltration, depending on the membranes used, and is performed in parallel to the membrane and through a pressure difference, allowing smaller volume components to pass through the filter while larger volume ones circulate along the membrane, preventing membrane fouling. This allows for continuous flow and increased yield.

The product that passes through the filter is called permeate, while the one that continues its trajectory is called retentate, which is recycled back into the feed. The TFF equipment consists of two main modules. The first one is the filtration assembly, which includes a single-use plastic tank with a volume of 2 to 5 liters that contains the product to be filtered and a filtration membrane with a surface up to 0.7 m².

The bag is supported on a support where it is located by a load cell and has a conical bottom to prevent product loss. The plastic tank is fixed on a support and contains the tangential filter that purifies the biomolecules. Both the filter and the support are gamma irradiated beforehand.

 To give a better idea of what these components are, here are the dimensions and weights of each part: Firstly, there are three different sizes of eBAGs that are used as the filtration containers. eBAG 5L has a working volume of 5 L and weighs 0.1 kg. eBAG 10L has a working volume of 10 L and weighs 0.15 kg.

Finally, eBAG 20L has a working volume of 20 L and weighs 0.18 kg. The control tower, which is used to manage the equipment, does not have a specific volume or weight as it varies depending on the specific model used. However, the tower is estimated to weigh around 35 kg. The peristaltic pumps are designed to work seamlessly with the TFF equipment, providing reliable and precise flow control for the filtration process. These pumps have integrated tubing that can be easily replaced when necessary and are compatible with a variety of tubing materials such as silicone, PVC, and TPE. The variable speed mode of these pumps allows for precise control over the flow rate, which is essential for maintaining consistent filtration performance. The analog signal control enables operators to adjust the flow rate to suit the specific requirements of their application, while the Drivesure motor ensures reliable and accurate operation.
Is a four-piston pump used for processing applications in the food, beverage, and pharmaceutical industries. The pump operates in a speed range of 30 to 2400 rpm and can deliver flow rates ranging from 0.333 to 20 liters per minute, depending on the eccentricity of the shaft. The maximum output pressure of the pump is 4 bar, and it has a filling volume of 75 mL without connectors. The pump uses tri-clamp connectors that are 3/4″ in size, ensuring a secure and leak-free connection.
The eLAB Tangential Flow Filtration (TFF) system has been engineered with AUTO-CIP functionality and equipped with automatic valves, enabling the seamless execution of complete Clean-In-Place (CIP) procedures. Additionally, the system is designed for compatibility with autoclave sterilization for enhanced ease and efficiency.
The eBAG and flowkit assembly come with specific specifications that are crucial for filtration and separation processes. The eBAG 5L model, manufactured by TECNIC, is made of TECNIC 9101 material, and the flowkit is composed of Sani Tech Ultra 65 and C Flex 374 materials. These components are designed to withstand the harsh conditions of the filtration process and ensure optimal performance.
